Figure 1: PCR-SSCP patterns obtained by processing rifamipicin resistant M. tuberculosis clinical isolates [sample (lanes) 2 to 11] and rifampicin sensitive reference strain of M. tuberculosis [ATCC27294] lane 1 and lane 12. All single point substitution were reported earlier except the double mutation which were detected at codon position 526 and 531 in three rifampicin resistant strains(lane 8). 9,14 The finding of a three band SSCP pattern may reflect the presence of two possible conformation for one of the DNA strands or a reannealing band. The codon numbers corresponds to E.coli RNA polymerase amino acid position.[15]
